ALEXANDER DISEASE: RARE LEUKODYSTROPHY & FIRST DISEASE-MODIFYING THERAPY

The US FDA has approved Zanvastro (zilganersen), developed by Ionis Pharmaceuticals, as the first FDA-approved treatment and first disease-modifying therapy for Alexander disease in children and adults.

What is Alexander Disease?

  • It is an ultra-rare, progressive genetic neurological disorder and a type of leukodystrophy, affecting the brain’s white matter.
  • It is caused by mutations in the GFAP (glial fibrillary acidic protein) gene, leading to abnormal accumulation of GFAP protein and damage to astrocytes.
  • It can cause progressive motor and cognitive impairment, speech and swallowing difficulties, and other neurological problems.
  • Historically, treatment has mainly been supportive, addressing symptoms rather than the underlying disease mechanism.

Zanvastro (Zilganersen)

  • It is an RNA-targeted antisense oligonucleotide (ASO) therapy.
  • It works by reducing production of GFAP, thereby targeting the underlying molecular mechanism of the disease.
  • It is administered by injection into the spinal canal (intrathecal administration) at regular intervals.
  • In clinical evaluation, treated patients demonstrated significantly better walking speed/motor outcomes than untreated controls.

  • It has a 1.05-million-token context window and supports up to 128,000 output tokens.
  • It demonstrates advanced capabilities in software engineering, science, computer use and cybersecurity.
  • Astra is the first OpenAI model classified at the “Critical” level for cybersecurity under its Preparedness Framework, reflecting its ability to identify previously unknown vulnerabilities and develop exploits.

STEGODYPHUS SARASINORUM: SOCIAL SPIDER & VENOM-BASED DRUG DISCOVERY

Why in News?

Indian researchers have produced the first comprehensive molecular and functional profile of the venom gland of Stegodyphus sarasinorum, identifying biomolecules with potential applications in anti-cancer and antimicrobial drug research. The study was published in Scientific Reports.

About Stegodyphus sarasinorum

  • Commonly called the Indian cooperative/social spider.
  • It is a colonial, permanently social spider found across the Indian subcontinent, particularly in semi-arid regions of central and southern India.
  • Unlike most spiders, it lives in permanent colonies, builds communal silk nests and hunts and feeds cooperatively.
  • Colonies are predominantly female, with males being smaller and darker.
  • It belongs to the family Eresidae.

Key Research Findings

Researchers used transcriptomics, proteomics, metabolomics and Confocal Raman spectroscopy to characterise the venom gland. The study identified 31 protein components, 32 proteins and 81 metabolites. Venom-gland extracts showed dose-dependent cytotoxicity against Dalton’s lymphoma ascites (DLA) cells, indicating preliminary anti-cancer potential. Certain venom peptides also have potential antimicrobial applications.

AGENTIC AI SKILLING PROGRAM: PREPARING INDIA’S WORKFORCE FOR THE AI ERA

The National Institute of Electronics and Information Technology (NIELIT) under MeitY, in collaboration with Intel India, launched the NIELIT–Intel India Agentic AI Skilling Initiative in September 2026 to expand access to industry-aligned AI learning.

Key Components

The initiative has two programmes:

  • Agentic AI for Everyone: Covers workflow automation, AI agents and multi-agent systems, with hands-on learning through no-code platforms. It focuses on productivity, business transformation and intelligent decision-making.
  • Engineering Agentic AI Systems: Focuses on designing, building and deploying agentic AI systems. It covers agent architecture, tool integration, orchestration, memory and deployment, using no-code/low-code platforms and coding frameworks.

What is Agentic AI?

Unlike conventional AI systems that primarily generate responses or analyse information, agentic AI can pursue goals, plan multi-step tasks and take actions with limited human intervention. Its core elements include perception, reasoning, planning, action and reflection.

Introduction

The formation of linguistic states transformed India’s federal structure. Potti Sriramulu’s sacrifice led to Andhra State (1953), followed by the States Reorganisation Act (1956) and the creation of Maharashtra and Gujarat (1960). The process showed that accommodating linguistic diversity could strengthen, rather than weaken, national unity.

 

The Andhra Movement and the First Linguistic State

The Fast and Its Aftermath

  • Potti Sriramulu: Began a fast unto death on October 19, 1952, demanding a separate state for Telugu-speaking districts of Madras.
  • Nehru’s Initial Stance: He was “totally unmoved” and ignored the fast, but soon acknowledged the Andhra dispute as a “festering sore”.
  • December 9, 1952: Nehru announced the government’s willingness to create Andhra from uncontested Telugu-speaking areas (excluding Madras city).
  • Sriramulu’s Death: Died on December 15, 1952 (58th day); hartals and violence followed.
